Open %CommonProgramFiles%\Microsoft Shared\Web Server Extensions\16\ISAPI\SharePointEmailws.asmx > Properties > Security (or run Get-Acl on it). Verify only WSS_RESTRICTED_WPG (the farm account) has Modify/Write access to the file.
